Alpha and Omega. Awal dan akhir. Ungkapan kuno itu kini diwakili oleh sosok Kate (Hayden Panettiere) dan Humphrey (Justin Long) yang memang bertolak belakang hampir dalam segala hal. Karena keadaan juga Kate dan Humphrey harus menyisihkan perbedaan yang ada ini untuk mencapai tujuan yang jauh lebih penting: selamat sampai di rumah.
Kate adalah seekor serigala yang selalu memegang teguh tanggung jawab dan disiplin dalam segala hal. Kate tahu kalau hidup di alam bebas penuh dengan tantangan dan Kate tak mau berbuat ceroboh yang nantinya bakal mencelakakan dirinya sendiri atau serigala lain dalam kelompoknya. Berbeda dengan Kate, Humphrey adalah seekor serigala yang hanya hidup untuk bersenang-senang.
Suatu ketika, Kate dan Humphrey ditangkap oleh seorang pemburu dan dikirim ke tempat yang sangat jauh dari tempat asal mereka. Berkat kecerdikan Kate, keduanya dapat meloloskan diri dari kandang namun masalah tidak berakhir di sana. Kate dan Humphrey harus mencari jalan untuk pulang kembali ke tempat asal mereka. Dalam perjalanan panjang ini mau tak mau segala perbedaan harus disisihkan untuk tujuan yang lebih penting ini.
